Two related biological problems are of enduring interest to our research group: What mechanisms at the subcellular and biochemical level are necessary to ensure normal function at cold cell temperature? What unique physiological and biochemical characteristics are expressed when species evolve under conditions of chronically cold body temperature? We have addressed the former question by experiments with both temperate zone fishes that experience cold temperatures seasonally and southern polar species that spend their entire life history at severely cold temperature. Our work with Antarctic fishes has addressed the latter question. For over two decades, we have been conducting field work on a regular basis in the area of the Antarctic Peninsula at the U.S. research base, Palmer Station, and aboard the research vessels that support this base. Fishes are the primary focus of our experiments because (with a few notable exceptions) they are nearly ideal ectotherms - i.e. their core body temperature is determined directly by the temperature of the medium that they breathe and in which they swim, water. For these animals, the \"environment-organism interface\" in the case of temperature, is directly at the molecular level. Indeed, cold temperature places a number of severe constraints on physiological and biochemical systems. These include reduction in rates of reaction, increases in fluid viscosity (of both body fluids and the medium), reduction in rates of molecular diffusion, effects on membrane biophysical characteristics and, in the most severe cases, threat of freezing. Our recent work has focused upon the Family Channichthyidae (Antarctic icefishes), which are unique among all vertebrate animals in lacking circulating hemoglobin in red blood cells; their blood is colorless. We have been examining the physiological and biochemical characteristics that allow the animals to compensate for the lack of an oxygen-binding protein in their blood and the evolutionary path that has led to this condition. Although most of our experiments are conducted at the subcellular/biochemical level, the underlying questions that guide us are firmly grounded in problems of \"real world biology\" at the organismal level. Ours is a research group that demands mastery of techniques as diverse as deployment of fishing gear at sea, protein purification and enzymology, and even current techniques in molecular biology.
